1 liter makes ~ 40 plates.
| 2X-ura-trp broth | 500ml |
| 5-FOA(fluoro-orotic acid) | 1gm |
| 20mM uracil supplement | 10ml |
| dextrose(glucose) | 20gm |
Place in 1 liter Erlenmeyer flask with stir bar. Heat on setting 2 or 3 while stirring. Heat is needed to dissolve FOA. Filter sterilize.
Melt 2 bottles of 250ml of 4% agar.
Add 250ml of 5-FOA media to each bottle of 4% agar.
Mix well by rotating bottle. Do not shake...this creates bubbles.
Pour plates.
