Ingredients for yeast
#1:
YNB w/o a.a.'s, amm. sulf. 8.5 g
Ammonium sulfate 25 g
-arginine drop out mix 10 g
Distilled water 2000ml
 
#2:
Dextrose 100 g
Distilled water 500 ml
 
#3:
Difco agar 100 g
Distilled water 2500 ml

Directions: Autoclave #1,2,3, separately (#3 with stir bar). After autoclaving, combine #1,2 with 3 and mix well. Stir at room temp. until flask can be handled without gloves. Add 15ml of a 20mg/ml canavanine stock soln. (Final conc. of canavanine = 60ug/L.). Mix well. Pour 25ml per plate. Canavanine sulfate is typically prepared as a filter-sterilized (0.2um filter) 20mg/ml stock solution in distilled water. It is stored at 4°C
